C013-99 The prolamins of sorghum, Coix and millets Leite A, Cord Neto G, Vettore AL, Yunes JA and Arruda P* Besides maize, other tropical cereals such as sorghum, Coix and millets should also be considered as interesting resources for seed protein studies. Sorghum was originally domesticated in Africa south of Sahara in ancient times and subsequently spread into other tropical regions. In the FAO report for 1994, sorghum was ranked fifth among the most important cereal crops of the world after wheat, rice, maize and barley, in both total planted area and production. Millets include various grasses species grown as food and forage crops in Africa and India and as high quality forage crops in the USA, South America and Australia. The origins of millet species are very diverse. Echinochloa millets were domesticated in Japan and Southwest Asia some 4,000 years ago. Pearl millet (Pennisetum glaucum) was probably domesticated 5000 years ago in the savanna south of the Sahara and west of the Nile, subsequently spreading to India some 3,000 years ago. Tef (Eragrostis tef), was domesticated in Ethiopia where it remains as an important crop until present times. Coix, commonly known as Job's tears, is a wild grass native to India, Burma, China, and Malaysia. It was grown extensively in South Asia before maize became popular as an agricultural crop. The Coix grain has been used as a food source for humans and livestock, but it's use declined because its agronomic performance competes poorly with maize and sorghum. P132-99 Plant uncoupling mitochondrial protein activity in mitochondria isolated from tomatoes at different stages of ripening Costa ADT, Nantes IL, Jezek P, Leite A, Arruda P* and Vercesi AE We have observed a higher state of coupling in respiring mitochondria isolated from green as compared to red tomatoes. Green tomato mitochondria produced a membrane potential (Delta Psi) high enough to phosphorylate ADP whereas in red tomato mitochondria, BSA and ATP were required to restore Delta Psi to the level of that obtained with green tomato mitochondria. This suggests that such uncoupling in red tomato mitochondria is mediated by a plant uncoupling mitochondrial protein (PUMP). Nevertheless, there is evidence that PUMP is also present in green tomatoes. It is suggested that the higher concentration of free fatty acids (PUMP substrates) in red tomatoes could explain the lower coupling state in mitochondria isolated from these fruits.. P133-99 The DapA gene encoding the lysine biosynthetic enzyme dihydrodipicolinate synthase from Coix lacryma-jobi: cloning, characterization, and expression analysis Dante RA, Cord Neto G, Leite A, Yunes Já, Arruda P* Dihydrodipicolinate synthase (DHPS) is the main enzyme of a specific branch of the aspartate pathway leading to lysine biosynthesis in higher plants. We have cloned and characterized the DHPS-encoding DapA gene from the maize-related grass Coix lacryma-jobi. The DapA open reading frame is interrupted by two introns and encodes the 326 amino acid-long Coix DHPS protein, which is 95% identical to the maize DHPS protein. Steady-state levels of DHPS mRNA were slightly reduced in the endosperms and embryos of the maize lysine-rich opaque2 mutants when compared with those in normal kernels. Selective binding assay with the maize Opaque2 protein (O2) showed that the GCN4-like element is not an O2 binding site, suggesting that the DHPS gene is not under the control of O2. P134-99 Implantes de colágeno liofilizado e gel de colágeno fibroso: análise histológica em ratos Palomari ET*, Vizzioli MR, Mendes Luz MA Foi analisada a reabsorção de colágeno liofilizado (CL) e gel de colágeno (GC) em dois diferentes tecidos. Fragmentos de 1 cm² de CL foram implantados no tecido subcutâneo dorsal de um grupo de ratos e outro grupo recebeu uma única injeção intramuscular de 1 ml de GC. Os grupos foram sacrificados após 3, 7, 11, 14, 17 e 20 dias. Fragmentos foram fixados em formol a 10%, incluídos em parafina e corados com H.E e T. Masson. Observou-se que o CL foi reabsorvido aos 17 dias, enquanto o GC ainda estava presente no músculo no 17° dia. P135-99 Ha-ras oncogene effect on DNA content and chromatin supraorganization in benzo[a]pyrene-transformed human breast epithelial cells Mello MLS*, Vidal BC*, Russo J When transfected to benzo[a]pyrene (BP)-transformed MCF-10F human breast epithelial cells (BP1 cell line) the c-Ha-ras oncogene has proven to enhance the neoplastic changes initiated by exposure to BP, giving rise to an aggressive tumorigenic cell line, BP1-Tras. We have previously demonstrated that BP afects the DNA content and the chromatin supraorganization of MCF-10F cells. Here, Feulgen-stained BP1-Tras cells were studied by image analysis in order to evaluate possible additional changes in DNA content and chromatin texture induced by insertion of the ras oncogene. A high variability in DNA content, and an increase in the packing states of the chromatin which became still condensed in BP1 cells were found in BP1-Tras cells. The results differed from those reported for the BP1-E1 cell line which is also an aggressive tumorigenic cell line, but was attained through progressive passages of BP-transformed cells. It was demonstrated that different patterns of changes in DNA content and chromatin organization may be involved in equally aggressive tumorigenic BP-transformed cell lines originated from the same cell line by different mechanisms. P136-99 Structural and histochemical observations on the hepatic system of Siphonops annulatus (Amphibia - Gymnophiona) Junqueira LCU*, Joazeiro PP The hepatic system of eleven specimens of routinely fed Siphonops annulatus was studied using light and electron microscopy, and histochemistry. lts general structure agrees with previously known information from amphibia. lts hepatocytes, however, present an unexpected scarcity of rough endoplasmic reticulum, few lysosomes and fat droplets, an abundance of glycogen beta particles and an exceptional amount of smooth endoplasmic reticulum. The macrophages, visible in its sinusoid capillaries (Kupffer cells), present morphological and histochemical characteristics that confirm their participation in the phagocytosis and digestion of aged erythrocytes. The abundance of leucocytes and occasional mitotic figures in the sinusoidal lumen suggest a possible intrahepatic hematopoietic activity. P137-99 Purification and kinetic properties of a castor bean seed acid phosphatase containing sulfhydryl groups. Granjeiro PA, Ferreira CV, Granjeiro JM, Taga EM, Aoyama H* An acid phosphatase was purified 2,000-fold to homogeneity, with a final specific activity of 3.8 µkat mg-1 protein from castor bean seed, through SP-Sephadex, DEAE-Sephadex, Sephacryl S-200 and Concanavalin A-Sepharose chromatographies. The relative molecular mass, and an apparent Km value were determined. Strong inhibitions by p-chloromercuribenzoate, Cu2+ and vanadate suggest the presence of sulfhydryl groups essential for catalysis. The castor bean enzyme also recognized tyrosine-phosphate and inorganic pyrophosphate (PPi) as substrate. A avaliação do padrão de distribuição espacial dos ninhos em uma colônia de 3500 indivíduos. Ninhos em 4800m2 da área da colônia foi mapeado. O fator mais importante para a escolha da árvore parece ser a presença de outros ninhos. Os guarás selecionam a estrutura do mangue principalmente pelo grau de distúrbio antrópico. P139-99 Geophagy in the golden-faced saki monkey, Pithecia pithecia chrysocephala, in the central Amazon Setz EZF*, Enzweiler J, Solferini VN, Amêndola MP, Berton RS A ingestão de solo de 9 cupinzeiros arborícolas foi observada em seis parauacus Pithecia pithecia chrysocephala (Cebidae, Primates; n= 853 h. obs.). Em março de 1987 ocorreram 25 eventos (19 dias, 132 h. obs.). A frequência não diferiu entre sexos. Os cupinzeiros apresentaram maior concentração de minerais que o solo. A capacidade de adsorção de taninos foi 10-20%, como no controle com caolin. A geofagia não ocorreu com alto consumo de folhas, nem sempre associada a frutos (reequilibrando minerais). A adsorção de taninos não é descartada, mas sendo um hábito irregular deve estar relacionado a componentes raros na dieta. Journal of Zoology (London) 247: 91-103, 1999. P141-99 Pharmacological evidence for *2-adrenoceptor in right atria from stressed female rats Spadari-Bratfisch RC*, Santos IN, Vanderlei LCM, Marcondes FK The purpose of this study was to demonstrate a physiological response to TA2005, a potent b2-adrenoceptors (b2-AR) selective agonist, in right atria isolated from stressed female rats under the influence of the estrous cycle. We obtained concentration-response curves to the agonist in the presence and in the absence of selective antagonists in right atria isolated from female rats submitted to three daily footshock sessions and sacrificed at estrus or diestrus. Our results showed that the pD2 values of TA2005 were not influenced by estrous cycle phase or foot-shock stress and confirmed a heterogenous b-AR population mediating the chronotropic response to catecholamines in right atria from foot-shock stressed female rats sacrificed at diestrus. The stress-induced response seems to be mediated by the b2-AR subtype. Right atria from rats sacrificed during estrus are protected against stress-induced alterations on the homogeneity of b-AR population. P142-99 Longitudinal differentiation in chromosomes of species of Sesbania Scop. Forni-Martins ER*, Guerra M Patterns of longitudinal chromosome differentiation were studied in five Sesbania species. After staining with fluorochromes chromomycin A3 and DAPI, each karyotype revealed two or four CMA+/DAPI - heterochromatic blocks, apparently corresponding to the nucleolus organizing regions. However, in S. virgata two or four CMA+bands were observed, depending on the population studied. No DAPI+ bands were observed. C-banding showed the most diversified patterns, being a more useful techinique to distinguish karyotypes of Sesbania species. P143-99 Interaction between ants, extrafloral nectaries, and insect herbivores in Neotropical coastal sand dunes: herbivore deterrence by visiting ants increases fruit set in Opuntia stricta (Cactaceae) Oliveira PS*, Rico-Gray V, Díaz-Castelazo C, Castillo-Guevara C This study examines the anti-herbivore effect of ants visiting the extrafloral nectaries (EFNs) of Opuntia stricta (Cactaceae), and its possible influence on the plant's reproductive output in Mexican coastal sand dunes. Opuntia's EFNs are located in the areoles of the developing tissue of emerging cladodes and flower buds. The associated ant assemblage was formed by 9 species distributed in 4 subfamilies, and the species composition of the principal ant visitors changed markedly from day to night period. Cladodes of control (ants present) and treatment (ants excluded) plants of Opuntia were equally infested by sucking bugs and mining dipterans. Damage to buds by a pyralid moth, however, was significantly higher on treatment than on control plants. This is the first demonstration of ant protection leading to increased fruit set in the Cactaceae under natural conditions. The results show how the association of EFNs with vulnerable reproductive plant organs can result in a direct ant-derived benefit to plant fitness. Functional Ecology 13 : 623-631, 1999. P144-99 Removal of seeds from vertebrate feces by ants: effects of seed species and deposition site Pizo MA, Oliveira PS* We investigated the removal of three seed species (Philodendron corcovadense and P. appendiculatum - Araceae, and Aechmea sp. - Bromeliaceae) from the faeces of Capuchin monkeys (Cebus apella) by ants in a lowland Atlantic rainforest in southeast Brazil. We asked if seed removal varied among seed species, between understory and gaps, and also among gaps differing in size and age. We recorded 17 ant species on the faeces, eight of which were observed removing seeds. On average, 68% of the seeds were removed from the fecal portions containing 15 seeds each after 24 h. For both the understory vs. gap and the among-gap comparisons seed species significantly affected the proportion of seeds removed, while deposition site had no effect. We interpreted these results as a consequence of the equalizing effect that three myrmicine ants had upon seed removal. Seed removal is likely to be less affected by differences in microclimatic conditions between understory and gaps than by the seed species involved. Canadian Journal of Zoology 77 : 1595-1602, 1999. P145-99 Ethylene and the inhibition of nodulation and nodule activity by nitrate in soybean Puiatti M, Sodek L* Thirteen species were analysed for nitrogen compounds in the xylem root bleeding sap. Amino acids were the main form of organic nitrogen found but only traces of ureides were present, whether the plants were nodulated or not. It was concluded that Crotalaria species have an asparagine-based metabolism, consistent with many other non-ureide-producing legumes. P146-99 Mineral nutrition and caffeine content in coffee leaves. Mazzafera P* The effect of nutrient supply on the caffeine content of coffee (Coffea arabica L.) leaves was investigated. Seeds were germinated in nutrient-agar media lacking N, P, K, Ca, Mg, S, Zn, B or Mo. The control treatment contained all essential nutrients. The caffeine concentration was determined seven months after seed sowing when the seedlings have 3 to 4 pair of leaves. The omission of K induced the highest caffeine content in the leaves (24.5 g kg -1). Caffeine in the control treatment was 21.9 g kg -1. Abscence of P induced the lowest content, 17.5 g kg -1. P148-99 Structure and ultrastructure of the spermatozoa of Bephratelloides pomorum (Fabricius)(Hymenoptera: Eurytomidae) Lino Neto J, Bao SN, Dolder H* The spermatozoa of B. pomorum are described at light and electron microscopy. As well as the spiraling of the nucleus, mitochondrial derivatives and axonemal microtubules, the sperm cells of this insect present other very different morphological features, such as those of the acrosome and centriolar adjunct, that differentiate them from the majority of the Hymenoptera. In adition, these structural variations show that the sperm of chalcidoids provide characters which can certainly be useful for future phylogenetic analysis at the subfamily level and, possibly, of the genus. P149-99 The functional value of sound and exploratory behaviour in detelencephalated pigeons Ferrari EAM*, Faleiros L, Cerutti SM, Oliveira AM Exploratory behaviour in response to a sound previously paired to a shock was analyzed before and after massive telencephalic lesions. Six pigeons were submitted to sound-shock associations, habituation tests, telencephalic lesion and habituation retesting. Eighteen birds divided into three sham-lesioned groups received sound-shock pairing (Control 1), pre-exposure to sound stimulation (Control 2) or pre-exposure to electric shocks (Control 3) before the behavioural tests. The data suggest a retardation in the typical reduction of responses to recurrent stimulation and support other evidences of the potentiating effect of sound-shock association on responding. They also indicate telencephalic modulation of exploratory behaviour and strenghten the arguments for the subtelencephalic storage of associative information concerning the functional value of acoustic stimulation.
